Collaborations
In order to complete the project better and help each other to move forward actively, this year we have collaborated with BNU-China, BS_United_China and DUT-China on plasmid construction, AHL-mediated quorum sensing system and model respectively. We also worked with LZU-China and NMU_China to explore how to popularize synthetic biology and coronavirus knowledge.
Collaboration with BNU-China

This year, we cooperated with the BNU-China team in the experiment. We provided them with a plasmid containing PmrC for experiment and shared what we have known about PmrC with them, especially in its special significance as a promoter. And BNU-China helped us to connect Hrp amplifier to the plasmids. We also exchanged ideas on plasmid construction experiments together.

Collaboration with BS_United_China

In the previous 2021 China iGEM Online Meetup, we have learnt that the project of BS_United_China also involved quorum sensing system mediated by AHL. Our project mainly used quorum sensing system, while the BS_United_China team's project mainly focused on monitoring AHL threshold, so our two teams had certain complementary roles in quorum sensing. Then our cooperation carried out.

We held a quorum sensing system online seminar together to share information about quorum sensing with each other. Then we started to help each other with the experiment. We received Part1 and Part2 of the engineered bacteria constructed by them for relevant experiments. The specific content of the experiment was to induce E. bsuahlscout to produce red fluorescent protein by using supernatant of the culture medium of engineering bacteria with different culture time. Overall, the experimental results were in line with our expectations, which helped us monitor AHL production efficiency and facilitated us to control a single variable to test AHL concentration gradient. The relationship between cultivation time and yield is shown in the figure below:


Testing AHL concentration in the environment by E. bsuahlscout helps us test our projects better. At the same time, we helped them conduct a simple evaluation of the effectiveness of their E. bsuahlscout.

Collaboration with DUT-China

This year, we collaborated with DUT-China in the model part. The team members of DUT-China took an example to explain the meaning of complex code in the model to us, and then we talked about the matters that needed attaching attention to in modeling.

In addition, we discussed the matters needing attention to about related genes in quorum sensing. For example, AHL degradation genes are necessary in eukaryotic cells, otherwise the cells will die. We also talked about searching LuxI homologous genes and the difference between the effect of each homologous gene. Through several working-togethers, both of us became familiar with the process of modeling, and attached more attention to the places that may lead to mistakes in modeling.

Collaboration with LZU-CHINA and NMU_China

We also collaborated with LZU-CHINA and NMU_China to discuss how to popularize the knowledge of synthetic biology and coronavirus. LZU-CHINA and NMU_China created the PPT for the presentation, and we contacted high schools and delivered the presentation. To make the presentation more interesting, we created short and interesting videos to help students understand. We also created a questionnaire for the students to research the effectiveness of our presentations, and the feedback was very helpful for all three teams.
