Overview
In this year's iGEM, the concept of "excellence in another field" was proposed. We don't just focus on working with hp, we think that iGEMers not only achieves our own achievements in science, but also thinks about how to influence the world in many ways and play a role of solidarity and collaboration. Combining science with aesthetics, we think this is a new requirement for human actual work.

So here's a demonstration of our work:

Crispr-Alliance
CRISPR, or Clustered Interval Short Echo Repeats, is an adaptive immune system for bacteria. In 2013, researchers applied CRISPR technology to eukaryotic cells to accurately and effectively edit genomes, opening a new chapter in gene editing technology. Since then, this technology has shocked the scientific community, CRISPR system in gene fixed-point editing, genome screening, gene transcription regulation, gene diagnosis and treatment and other fields of research and application began to blow out. In this year's project, we used crispr technology to knock out the FadR gene to enhance beta oxidation and help engineer bacteria to better metabolize fatty acids.

When we worked with CCiC, through communication with many teams, we learned that teams using CRISPR technology have issues that need to be addressed in terms of CRISPR technology.

Therefore, Sponsored and hosted by Tianjin, BUCT responded positively and assisted in contacting other teams to ensure the successful conduct of the exchange with ZJUT-China, OUC-China, NNU-China, NWU-CHINA-A, NEFU_China, XJTLU-China, CPU_CHINA, LZU-CHINA and Tongji_Software participating in the crispr conference.

We participated in a one-day CRISPR conference with 11 iGEM teams. The conference consists of project presentations, proposals, free discussions and expert sharing to address the technical challenges of using CRISPR for iGEMers. At the conference we shared the technical obstacles we encountered in using Crispr technology to knock down the fadR of the Nissle 1917 genome and received valuable advice from others on improvements in experimental operations.

In addition, we have co-written a CRISPR APPLICATION GUIDELINES, hoping to have some inspiration for future teams using crispr.

The intestinal project Alliance
On August 6, 2021, we co-hosted the Intestinal Conference with six teams, including SUSTech-China、SZU-China、NJMU-China、HZAU-China 、Nanjing-China and BNUZ-China. At the conference, we not only discussed our project, but also co-authored the 2021 Intestinal Project Reference Manual. The protocol consists of several parts, of which we are responsible for writing the intestinal value section.

As we all know, intestinal-related projects often involve the planting of engineered bacteria, and the formation of healthy intestinal flora plays an important role in the formation of immunity and the establishment of intestinal barrier function. Probiotics eventually work at the lower end of the digestive tract, the lower part of the small intestine and the large intestine, so whether probiotics can fight various digestive fluids in the digestive tract to reach their destination - the intestinal wall depends on their ability to "stick" and "plant". To this end, our team has prepared practical cases of previous iGME teams for introducing engineered bacteria into the body and adopting a planting strategy, hoping to provide reference for subsequent iGEM participating teams.
In addition, we have co-written a CRISPR APPLICATION GUIDELINES, hoping to have some inspiration for future teams using crispr.

Artistic creation
After amplifying individual genes and plasmid pCS and pSA through PCR technology, we constructed pCS-lac-trpB-pcd-p4h-T1 and pSA-lac-trpE-trpG-AntrpC-T1 by enzymatic slicing, respectively, and identified two plasmids by enzymatic slicing. The two plasmids were then converted into E.coli Nissle 1917, cultured at a constant temperature of 12h at 37 degrees C, and then inoculated into a shaker bottle with three colonies, fermented at 37 degrees C with glycerin as a carbon source, sampled every 12h, fermented 60h. After the sample was made, the 5-HTP of the fermented sample was measured in liquid phase, and the data results were as follows:
In the process of doing the project, we thought about the issue of bodyshame. Therefore, our team invited Jiaxin Yi to write a beautiful song "free me from the cage", which was sung by Jiaxin Yi and Qilin Zhao. We filmed a movie and published it on bilibili(), which get a lot of affection and approve on the Internet. Through this song, we expressed our attitude and clarified the intention of our project. We are to help those who are deeply overworked and depressed to solve their difficulties, not to sell weight-loss drugs to promote body anxiety. We hope that the public have the correct cognition and dare to break free from the shackles of body anxiety and pursue true health.

The composer Jiaxin Yi said: "This is a song about bodyshame. This is the first time I have written a song to think about and feel these troubled groups. I have seen some People who experience bodyshame bravely express themselves, they do not succumb to the injustice or even bullying caused by people’s stereotypes of “beauty” in reality. At this moment, regardless of their appearance and body type, they are the world. The most beautiful and shining people in the world. Thank them for their efforts to make the world more tolerant, so that more people can discover and understand the'beauty' from different perspectives of themselves, others, and the world."
