Although our planet provides us with an abundance of water, we can only utilize roughly 2,7% of drinking water. That leaves 7.8 billion people with access to only 0.4 % of the water resources. Many people suffer from this shortage and with a rapidly growing world population, it is bound to get worse if changes will not be implemented. Moreover, countries like Sweden, where we are located, have never had to think about a lack of accessible water so far. But due to the climate crisis also those countries will face droughts in the future.

Additionally, current desalination methods are highly energy and resource-consuming.

Our solution to this dilemma is the light-driven desalination of water by halophilic cyanobacteria which possess the photosynthetic ability, in order to tap into the vast resources of the oceans. We will genetically engineer Synechosystis sp. PCC 6803 to express the inward-directed chloride pump Halorhodopsin in a light-dependent manner. Furthermore, we will genetically modify them to enhance a sodium channel expression that transports sodium ions into the cells following a gradient created by the chloride ion influx. By expressing these ion pumps together with the enzymes required for efficient ion transportation, desalination of seawater will be achieved.

Moreover, we want to present an easy solution for separating cyanobacteria from the desalinated water. To achieve this, we will modify them to express a fusion protein containing a membrane-anchored protein as well as a carbohydrate-binding domain on the outer membrane. This will allow an easy separation by filtering the water through a cellulose membrane leading to covalent binding of the bacteria to the cellulose.

Ultimately, we want to make our concept suitable for industrial-scale use as it has a clear advantage of being eco-friendly, by using sunlight instead of large amounts of electricity like current desalination methods.
