Preface

Security of project design: The microorganisms used in the project include EScherichia coli DH5α and Saccharomyces cerevisiae, which are harmless to other organisms and the environment. We hope to use these two strains to produce linalol and nerolol. In the preliminary work, we carried out an in-depth investigation on the relevant information of Linalool and neroli, and various literatures showed that the two spices were safe and harmless at the prescribed concentration. Linalool (S0029) and nerolol (S0046) are food additives approved for use in China's NATIONAL Food Safety Standard GB 2760-2014.

High concentrations of linalool and nerolol pose potential threats to the environment and organisms. In order to avoid the risks, we chose non-high concentration culture to control the yield of linalool and nerolol.

In the early laboratory work, we used plasmids with resistance genes for the screening of transgenes and transformants, but after testing, we transferred the target gene into the genome of the nutrition-deficient strain by homologous recombination. The nutritionally deficient strain can only survive in a special medium and will die soon after leaving the medium.

The main exogenous genes of the fragrance-producing strains were constructed from plants, and the relevant sequences were downloaded from NCBI and synthesized by the company. The proteins encoded by them were safe.

All members must pass the theoretical and operational test of experimental safety before entering the laboratory.

Our instructor, Zhang Qinghua, organized the lab safety training. Contents include: laboratory structure and infrastructure distribution location, the use of various experimental instruments and precautions, fire equipment, emergency ventilation and emergency shower devices,etc.

In the lab, we ask that all people have to do a good job in basic protective measures, (protective clothing, gloves, goggles, etc.) in strict accordance with the provisions and bacteria to deal with the rubbish in the biochemical culture medium, in strict accordance with the provisions of access chemicals, in strict accordance with the specification standard operating instruments, shall not be placed in laboratory work area life items, shall not be laboratory items to rest area.

There are students in the team who are responsible for lab management. They are responsible for bringing new members to be familiar with experimental operations until they can operate independently. In addition, students in charge of laboratory management will check the laboratory regularly and report to the instructor to ensure the order and safety of the laboratory. In addition, our advisor and the lab safety officer will also visit the lab from time to time.
