Yina Li - Wet team leader, in charge of monitoring the team progress and organizing tasks. Participation in lab process including culturing bacteria, plasmid extraction, purifying proteins, and conducting SDS-PAGE and agarose gel electrophoresis gel.

David Gao -Wetlab team member in charge of: preparation of liquid and solid LB culture medium, culturing bacteria containing the gene of interest in the culture medium, extracting PET28a-MHETase plasmid, preparing agarose gel for electrophoresis, adding samples to agarose gel and SDS-PAGE electrophoresis gel, selecting monoclonal bacteria colony and culture in the culture medium, making the agarose plate and coating the bacteria onto the surface to culture, preparing nickel column purification, overlap PCR and enzyme ligation, and testing OD600 value for the bacteria solution.

Alethea Huang -Wetlab team member in charge of the preparation of LB culture medium, transformation into BL21 competent cells, coating plates, selecting monoclonal colony PCR, extracting pET28a-MHPETase plasmid, samples loading to agarose gel and SDS electrophoresis, preparation of protein purification Buffer reagent A (25mM imidazole), nickel column purification, overlap PCR and Enzyme ligation, machines operation, such as shakers, water bath, and centrifuge, and experiment data recording for MHPETase group in Xuhui.

Tony Li -Wetlab team leader in charge of the coordination of tasks, monitoring experimental progress, sterilized LB culture medium preparation, plasmid extraction, plasmid endonuclease digestion, SDS-PAGE gel electrophoresis, DNA gel electrophoresis, BL21 transformation, bacteria colony coating, nickel column protein purification, PCR amplification of IsPETase and MHETase, DNA ligation, and enzyme & bacteria activity testing.

Shu Yang -senior student from Shanghai World Foreign Language Academy. Besides an interest in biological fields of genetics and biochemistry, I myself with interdisciplinary research between and beyond STEM courses.
