Integrated Human Practices
Human Practices work has deeply influenced the conception, design and stage of our project. We studied Plastic Package market up and down our project purpose. Our consultation with experts helps us design our projects. Feedback and experience from companies and governments help us design projects for use. Improvement of an Existing Part
This year, we tried to improve the degradation efficiency of PETase by improving the thermal stability of PETase. We’ve registered many improved parts of PETase this year. Among them, as an improvement of the existing part BBa_K2010000, BBa_K3715005 shows extraordinary thermostability. For more information, check out our Parts page.
Project Modeling
We built the Mathematical Modeling: UV-HPLC to simplify complex experimental operations. We also evaluated the thermal stability of mutants by Molecular Dynamic Simulations. Homology modeling was conducted using Alfold and affinity for ligands was calculated using Docking. Please visit Modeling pageto see more.

Education & Communication

Collaboration

Human Practices
The reason we chose this project was because of our concern for the environment, we further carried out some background retrieval. To ensure that our project is beneficial to the society, we conducted some social interviews, including enterprise research and government interviews. Please visit Human Practices pageto see more.
Proposed Implementation
Our target users are companies that produce, recycle and reuse PET products, waste treatment plants, environmental organizations and so on. We designed an industrial system that included the design of the fermentation tank and the processing process. Please visit Implementation pageto see more.

Project Description
Our aim is to improve the degradation rate of a novel PET degradation enzyme named PETase.  We tried to improve the thermal stability of PETase and finally improve the degradation efficiency of PETase. Please visit Description pageto see more.
Contribution
Our contributions for future iGEM teams are as follows:
  • Parts:Seven PETase mutants’ thermal stability were greatly improved
  • Found rules to stabilize protein
  • Constructed a testing method of a catabolic enzyme.
  • Established a method which can accurately predict protein structure
  • Developed a conversion model between UV260 and HPLC Peak Areas
