Contributions
CotA (BBa_K2684000) is derived from B.subtilis and a protein that will decolorize a wide range of dyes as the catalyst. For our contribution, we provide extra experiences of using this gene in the E. coli BL21 and get slightly different result with the experience listed before. Here is the following information: We figure out slightly different data with the previous group, even though we have used the same RR red color with the group before. The data are shown below, cotA+ means cotA added with iptg (got activated and expressed), and cotA means there is no iptg added. From this data, we can see that the color degradation is already clear during 24h, and in the next 24h, the absorbance barely decreases for cotA+. Therefore, we came to the conclusion that the color changing time will be around 12-24h to be more precise rather than 48hr.
In addition, we have done some modelling work to evaluate the degradation ability of this protein. More information can be views in https://2021.igem.org/Team:THIS-China/Model
