Abstract

Dental plaque is an ecological environment in which bacteria grow, reproduce, decay, and carry out a series of metabolic activities. In this project, an E. coli biosensor is built to activate the phosphorylation of ComE membrane protein and ComD protein in sensor cells and activate the downstream CotA gene to cause color degradation. This sensor cell uses short-length peptide CSP secreted by Streptococcus mutans as a signal to detect the amount of S. mutans in the patient's mouth. To demonstrate this, contactless cell-free system hardware is designed. The customers can use the oral rinse to collect the sample and place into the device to see color degradation. The product is portable and reasonably priced, thus allowing more customers to have the ability and willingness to purchase it.

Ben did work in the dry lab, supported our work in collaboration, and made a preliminary draft for the website.

Fatima Hachem: Primary Instructor

Ms. Hachem helped us put together the whole project. She was in charge of the payments and registration, and also gave us essential advice on different parts of our project.

Xuan Wang: Secondary Instructor

Xuan Wang taught us basic lab safety and lab techniques, and gave us essential advice for our experiment, HP, and collaboration.

Xiaofei Ge helped us research and set up our topic, built up the basic part of our iGEM project, and helped us on certain problems we met.

The Tsinghua team helped us settle down the project, solved some problems we met during the experiment, and communicated about HP, hardware, and website design. We also led promoting synthetic biology with them.

QHFZ and our team held joint meetings. We borrowed equipment from each other's labs, and checked each other’s projects.
