    2. From 2020 to the present, the COVID-19 pandemic has not faded, which makes people’s communication have undergone great changes. To slow the spread of the COVID-19 pandemic , most meetings and events are held online. But that didn’t stop us from working with other teams. We got to know the iBowu-China team through this year’s Northwest China online meeting, and the XJTU-China team through the offline meeting, which attracted dozens of teams to share and exchange projects!





      From May to October, we maintained some monthly exchanges, especially with the iBowu-China team, who was also targeting research in Chinese herbal medicine, targeting the improvement of herbal production, and we discovered similarities and directions for deeper collaboration between our two teams' projects. There were also several follow-up online meetings for discussion of the experimental process, mathematical modelling, poster design and the refinement and checking of safety forms. This has been a great boost and improvement to the progress of the project between our teams!


      Here are some of the details of our conversations with both teams.






    3. Project Communication and Human Practices Encouragement


    4. They wanted to make a promotional film on herbal medicine this year, so we provided them with information on our research into ginsenosides, which we hoped would be of some use in the documentary they were making. At the same time, they had ideas for the production of education derivative design solutions, and we exchanged many experimental ideas with each other, and also helped to contact some teams for the iBowu-China team who were interested in participating in as much collaboration as possible, in preparation for the iBowu-China team's post-production doodlebook project.

      Their team mailed us a product they use for education - herbal scented sachets, as a testimony to the friendship between the two teams. At the same time, their team shared with us the records and information from their HP interviews with herbal medicine companies, giving us an insight into the development and some of the research and development methods of the current pharmaceutical companies specializing in herbal medicine. It gave us another insight into the popularity of herbal medicine in the economic market and its potential development power, based on the human practices we have already done.

  •   Experimental help





  • Their team asked us about the operation of the HPLC liquid chromatograph that they were unfamiliar with, so we helped the iBowu-China team step by step and solved this problem. Our team members taught the iBowu-China team online + on-site in the lab and also shared our current product recordings using HPLC with their team. In addition to this, we shared the work that we have done regarding protein structure prediction, which has inspired their team by analysing the sites where genes can bind through protein structure prediction. At the same time, we have chosen to match our own project with the Pichia pastoris and we have used the AOX1 promoter, which better enhances the expression of the target gene, for their team.





    As the concentration of plasmids coming out of electrophoresis was too low, their team helped us to analyse the reasons for the failure of electrophoresis and speed up the progress of the experiment, while we exchanged the enzymes of the two teams and tried to catalyse our reaction substrate ginsenoside Rb1 by their enzymes, but it did not work as well as the enzyme we initially designed.







  •   What we do together



  • Together, we participated in the 2021iEEE International Conference to educate the general public and popularise the science of synthetic biology, to introduce young people and scholars from other research fields to the wide impact of synthetic biology in the world, and to present the projects of our two teams. The response from the general public has been very positive and new to the field of synthetic biology, and the educational outreach by our two teams has resulted in a group of young people who are intensely curious about the synthetic biology community and we encourage them to continue their exploration.

  • Project Communication and Human Practice Encouragement. Their team's research is on the microbial synthesis of tryptophan for improving human sleep problems, and ours is on the transformation of ginsenosides. Both teams have production-related aspects in common and encourage each other to find relevant companies for human practice.








  •   Experimental help





  • The cell we used was Pichia pastoris, and we explained to them exactly what we thought about the experiment, while we learned that their team wanted to finally confirm the results of the project by measuring the yield. We recommended measurement by HPLC and offered help with HPLC. In the follow-up communication, they had problems with positive control, a sequencing situation and without any product after gum-cutting recovery, and we recommended Shanghai Shenggong Bioengineering Technology Service Co.,LTD.





    Their team used a leaky promoter with low inducible expression, used fitz repressor protein to hinder bacterial division to regulate growth, and on this basis again reduced bacterial division again by selecting a leaky promoter with low expression to improve the operability of later experiments. This inspired us to use the AOX1 promoter and to link methanol to the AOX1 promoter. In response to our plasmid concentration being too low, they suggested that we increase the culture volume, improve centrifuge tube utilisation and ensure that the concentration increases after electrophoretic recovery, to avoid too low a concentration of plasmid transformation leading to unremarkable experimental results. Their team mentor introduced us to the fact that crude enzymes derived from Lactobacillus paralimentary LH4 can also convert ginsenoside Rb1 to CK, broadening our horizons in the field of ginsenoside conversion.
