We believe that the problem of land pollution and global warming brought by fast fashion should be a public concern. Our project aims at providing an alternative material of cotton and other clothing material for the clothing industry to use. Upon completion of the tailor-made fermenter, the mass production stage will be introduced. We sincerely hope that this biosynthetic fibroin will help in saving our mother nature.

Upon the wiki freeze, we have found different types of silk fibroin, namely major ampullate spidroin, tubuliform and pyriform. Each type of silk has different properties as spiders naturally use different silks in different situations, therefore to attain the best silk we have to find a way to do proportional expression. However, that is our long term goal. In this year we focused on the successful transformation of plasmid containing different types of silk into the 2 types of E-coli namely DH5𝜶 and BL21.

Besides, our team produces a standard curve for IPTG and arabinose respectively by using our modelling data. With the help of these two standard curves, we are able to find out how much inducer to be added to produce a certain amount of protein. Moreover, the ratio between the amount and inducer and concentration of protein produced has been significantly shown on the graph. We feel glad to share these information with all iGEM teams and hope that these information is helpful to them, so that they can make use of these data in their future work.

Despite the countless errors and limitations in our project, we enjoyed different limitations and the processes of finding solutions to them. We appreciated the help and genuine support from other iGEM teams and experts in the field. We truly believe that the difficulties we have will polish our project and finally turn stone into gold.
