With Thinker-EDU-China & LZU-HS-China

In June, we communicated with our instructor and got in touch with another 2 high-school IGEM teams. We, LZU-HS-China, Think-Edu, XHD-Wuhan-Pro-China, are in the same track and we have similar ideas about our projects. We formed a high-school league to promote further collaborations. We also invited LZU to be our undergraduate and graduate mentor. We invited other school teams and bioers to join our league and held forums online twice a month. We discussed potential problems within capacious domains under tight collaborations throughout the whole iGEM season.

1.We recommended LZU-HS-China whose goal was also to construct an intestinal probiotic to use E.coli Nissle 1917 as the chassis creature due to its high safety and highly available genetic information. the genetic background of common probiotics is unclear and difficult to engineer. The common e. coli has internal toxicity and is not suitable for human or animal intestinal function. So we used 1917, which had the advantage of being engineered without endotoxicity. We tried 1917 and it worked, which gave other teams the insurance, confidence and experience to use 1917.

2.We discussed the activity of conducting expert interviews and hospital field investigations together. After the discussion, we decided to go to the hospital together for an interview, and we also discussed the question design and the contents of the hospital investigation.

3.While we were making our project promotion videos, we encountered a technical issue—The mechanisms behind our project are difficult to be explained thoroughly using simple graphics. We decided to make a short, explanatory animation that could be useful for developing a clear structured and well-developed video. However, our art group was not familiar with computer software. Fortunately, we were able to obtain help from LZU-HS-China. They guided our team members to create short animation clips for our videos using Adobe Aftereffects and Adobe Premiere. It helped us to clearly demonstrate the process of alcohol digestion and the functions of our product.

4.We decided to help promote each other's projects in our own schools, so as to jointly promote the development and communication of each other's projects.

5. Members from LZU-HS-China discovered that because EcN contains secret plasmids, it can interfere with the expression of foreign genes at specific locations to some extent and disturbed the efficacy of the gene and protein. Thus, they ameliorated their project by adopting a new high copy plasmid-pSB1A3. Moreover, the strong promoter before the expressive sequence and INP, an appropraite and stable protein transporter binding on cell membrane, were added in front of the target gene to promote the efficiency and stability.LZU-HS-China shared these achievements with us and helped us avoid potential deficiencies in our experiments.
