Overview

Our part collection consists of toehold switch biosensors aimed at detecting the plant pathogens Fusarium oxysporum and Phytophthora cryptogea. These were designed by finding unique genes required for pathogenicity in both species: FRP1 for Fusarium oxysporum and X24 for Phytophthora cryptogea. Each toehold was composed of a T7 promoter, the respective toehold sequence, GFP, and the T7 terminator. Each trigger was composed of a T7 promoter, the corresponding trigger sequence, and the T7 terminator. In the presence of the trigger sequences, the hairpin loops in the corresponding toehold parts would unravel and allow for expression of the downstream GFP.
