Safety
The general safety of our laboratory
In the 12-day training, safety instructions are indispensable for ones who
frequently do experiments in the laboratory. On the first day of entering the laboratory, our supervisor
gave a lesson about laboratory safety. First, laboratory clothes must be worn neatly, which helps protect us
from being harmed by hazardous substances during experiments. Second, long hair must be tied or arranged, so
that the solitary hair will not influence experiment progress or shield eyesight. Third, food and beverage
are prohibited from entering the laboratory which prevents the experimental area from been polluted by them.
After finishing the experiment, hands must be washed before leaving. Fourth, laboratory equipment, such as
microwaves and refrigerators, are not allowed to preserve or put an impact on food. Fifth, do not touch
electrical equipment with wet hands. Sixth, keep the surroundings nice and clean, and strictly differentiate
the experimental and clean areas. Seventh, prepare equipment needed for later experiments and review the
process. Eighth, all experimental wastes should be sorted and carefully treated before abandoning. Due to
the epidemic, the school started to take our temperature every morning to make sure that we have no evidence
of having a fever.
Material Safety
The microorganisms and experimental equipment we used in the experiment operation:
The plasmid used are pUC57-amilGFP and pUC57-Pprovoin5. Regarding the microorganisms, we only used E. coli
which is commonly used and all the related experiments were conducted inside the laboratory under
supervision only. Ethidium bromide, EB This used to be what is added to the electrophoresis gel, but
considering the strong mutagenicity and toxicity, we replaced it with a safer nucleic acid dye. Autoclave
Check the liquid level of the autoclave before using. Tap water should not be used or the components will be
rusted. When the temperature is too high, the valve gate should be closed. An ultraviolet platform is used
to observe the DNA stripes on the electrophoresis gel. When using it, do not stare at the light for too long
in case it causes discomfort to the eyes. We use an alcohol lamp for high-temperature disinfection. Pay
attention when moving the lamp, use both hands and do it slowly. Use the lamp cap to extinguish the fire
instead of using other ways, such as blowing with your mouth.
