  • Butanol production in PCC 11801 (wangikar's strain, isolated from Powai lake, beside IIT Bombay)
  • They started working on iGEM in late Aug, 2020, they decided on doing something with sustainability and cyanobacteria in Jan '21
  • Goméz had done iGEM in 2018? or 2019?
  • They have lab access from June to September
  • They are interested in PCC 11801 because it's:
    • robust
    • naturally transformable
    • grows as fast as UTEX 2973 even under ambient conditions, fast growth is a basal phenotype and not something that kicks in at higher temperatures, light intensities and CO2 concentrations, and they want to have open air cultures in their industrial set up
  • They want to develop tools, find specific sites, optimise growth parameters and generally characterise PCC 11801
  • They have filled out all the necessary forms and declarations and waiting to get approved by some Indian govt body so that they can acquire the PCC 11801 strain, they are quite adamant to use it, and will wait till June to see if they can get their hands on it, otherwise they'll switch to a different strain.
  • They were interested in a consortia set up (especially with Yeast) but they were particularly worried about managing the growth rates of cyanobacteria and heterotroph
  • They wanted to explore possible mixotrophy to allow their cyanobacteria to feed on sucrose at night time in order to ensure continual production of butanol at night and even in winter or gloomy days. The sucrose would be generated by sucrose secreting cyanobacteria in another tank. They aren't sure whether additional engineering done on the cyanobacteria to make them mixotrophic would lower their productivity, thus counteracting any benefits obtained from night time production.
  • They thought we could assist them in modelling for the mixotrophy bit becuase we will be working on sucrose secretion and consumption via cscB (Dry Lab should speak to them)
  • PCC 11801 doesnt have a characterised fluxome out yet and so they plan to use a top down approach of making use of the genome of PCC 11801 and comparing it to the genomes of other related cyanobacteria whose fluxome is known in order to guess the constraints for their FBA modelling
  • They developed a programme that can find potential and non-essential neutral sites in any genome
